Heparan Sulfate Proteoglycans

Heparan sulfate proteoglycans (HSPGs) are found at the cell surface and in the extracellular matrix, where they interact with a plethora of ligands. HSPGs are glycoproteins, with the common characteristic of containing one or more covalently attached heparan sulfate (HS) chains, a type of glycosaminoglycan (GAG). Cells elaborate a relatively small set of HSPGs (~17) that fall into three groups according to their location: membrane HSPGs, such as syndecans and glycosylphosphatidylinositol-anchored proteoglycans (glypicans), the secreted extracellular matrix HSPGs (agrin, perlecan, type XVIII collagen), and the secretory vesicle proteoglycan, serglycin. HS is evolutionarily ancient, and its composition has remained relatively constant from Hydra to humans (see Table 1).

Chondroitin Sulfate Proteoglycans

Chondroitin sulfate proteoglycans (CSPGs) consist of a protein core on which there are one or more serine residues that can act as the sites of attachment of glycan chains. They are used as barrier-forming molecules in many biological events. Various observations lead to the conclusion that they are important inhibitory molecules in the reactive glial tissue surrounding injuries. Several CSPGs, including neurocan, phosphacan, versican and aggrecan (see Table 2) are upregulated in CNS scar tissue, all of which can inhibit axon regeneration. Therefore, Chondroitinase ABC enzyme (purified from Proteus vulgaris) degrades these CSPGs and has been shown to promote functional recovery and neural regeneration in addition to its role as a tool in glycoanalysis.

Keratan Sulfate Proteoglycans

Keratan sulfate proteoglycans (KSPGs) play a pivotal role in the development and maintenance of corneal transparency. Keratocan, lumican, and mimecan (osteoglycin) are the major KSPGs in vertebrate corneas (see Table 3).
