Exosomes are small endosome derived lipid nanoparticles, actively secreted by exocytosis in most living cells. Acting on tissues in both the local environment and further afield, they function as important signal carriers through their transport of RNA, proteins, lipids and DNA. Exosomes are involved in a wide range of normal and pathogenic processes, making exosome research crucial in multiple areas. Our Services

Exosome Isolation & Quantification

Our Exosome Isolation & Quantification Services can reliably and reproducibly isolate exosomes from almost any biofluid—from plasma and tissue culture media to cerebrospinal fluid. Exosomes will be isolated by ultracentrifugation and quality will be assessed using a NanoDrop spectrophotometer.

Options are also available to isolate by differential/gradient ultracentrifugation, immunoaffinity, size exclusion chromatography, precipitation/low speed centrifugation.

Exosome miRNA Isolation & Sequencing

For our Exosome miRNA isolation and sequencing service, we will extract RNA from exosomes, and subject it to the Small RNA-Seq (Illumina Platform). Basic analysis will be performed on the raw data. Screening for specific miRNA can also be performed.

Exosome Surface Marker Analysis and Proteomics Service

AMSBIO offers analysis of exosome surface proteins using MACSPlex kit. For proteomics analysis, samples will be analysed by Agilent 6530 LC/Q-TOF mass spectrometer to characterize the peptides formed. The analyzed samples will be differentially analyzed using molecular features finder and mass profiler (Agilent software) to determine changes in the peptide profile.

The MS/MS data from the selected peptides will be imported into Spectrum Mill proteomic server (Agilent software) to identify the protein that is consistent with the imported peptide sequence. The NCBInr and Swiss-Prot data bases are available to search.

Exosome-induced cell proliferation

      • Responding cell types: Human Keratinocytes or Human Fibroblasts
      • Comparison to a maximum of 5 known inducers
      • Includes serum and non-serum control media
      • Cell Titer Blue Assay for cell proliferation

Exosome-induced collagen production

      • Responding cell types: Human Keratinocytes or Human Fibroblasts
      • Comparison to TGF-β1, a known inducer
      • Includes serum and non-serum control medias
      • Tested using ELISA for pro-collagen 1 C peptide
      • Total Cost: Dependent upon number of samples, titrations requested, and cell types tested

Exosome-induced inhibition of inflammatory cytokines

      • Responding cell types: Human Keratinocytes or Human Fibroblasts
      • Comparison to lyophilized heat-killed bacteria, a known inducer
      • Includes serum and non-serum control medias
      • Tested using RT-qPCR for IL-6, IL-8, and MCP-1
      • Total Cost: Dependent upon number of samples, titrations requested, and cell types tested

In vitro wound healing (scratch assay)

      • Standard scratch assay will be performed using monolayered cells in each well, with photomicrographs taken at 12 hr intervals over a 72 hr period
