Gold-in-a-Box™ Colloidal-Gold Conjugation Kits
For preparing highly reactive antibody (purified) and protein (purified and soluble) gold conjugates
AMSBIO offers the Gold-in-a-Box™ kit, which enables users to quickly determine (in less than one hour) the optimal colloidal-gold binding conditions for attaching soluble proteins, and the Fc portion of purified monoclonal and polyclonal antibodies. The resulting gold particles are highly active and greatly increase the sensitivity of lateral-flow tests. Customized components are available upon request.
Advantages
- Uses consumables that are pre-validated eliminating the risk of purchasing non-compatible reagents, saving troubleshooting
- Significantly reduced antibody costs when compared to physisorption
- Save time due to reduced number of conjugations needed to determine optimal colloidal-gold binding conditions (within an hour)
Bioassay Works range is based on gold nanoparticles so small they appear intensely red and remain suspended in water making them ideal for rapid, point of care (POC) assays to detect minute traces of important substances.
To accomplish this feat, nano-gold particles are coated with appropriate receptors that react specifically with the molecules to be detected.

Background
Lateral-flow chromatographic and flow-through assays offer fast detection of critical components for use in point-of-care testing. The key to flow-through tests is the ability to covalently attach antibodies to intensely colored, nanometer particles.
Gold sols that bind ligands through a sulfur bond have proved highly successful for this application. For optimal binding of the antibody or protein while retaining a high degree of specific activity, the pH of the gold sol must be adjusted to slightly above the iso-electric point of the coating antibody or protein.
This is done through a series of pH titrations with the provided buffers. Varying amounts of buffers A and B, and varying amounts of buffers C and D are added to the gold to create a pH 5-11 range. Next, antibody or protein is added, and after 30 minutes the reaction is stopped.
